Server Connection Interrupted
I see there is a history of issues with SMB connection on Mac OS. These seem to have been cured in January 2017 for the most part. But I wonder if the errors are back with 4.3.3.0299 9/1/2017?
I just set up QNAP TS-451+ yesterday and simply trying a time machine backup. There are great directions here:

No problem following these instructions and mounting... TM runs and I got at least 20GB into it once but then the backup fails and Time Machine preferences displays message like Waiting for first backup.
I then noticed the "Server Connection Interrupted" errors so I am guess the two are related and TM is failing due to a Server Connection Interruption.
Can anyone confirm 4.3.3.0299 is working for their TM backups?
Anyone have other ideas why I'm getting Server Connection Interruption errors?

Server Connection Interrupted
Make sure your SMB version is set at 2.1 in Qnap. Also make sure you have disabled smb signing on Mac side.

Thanks. Removed disk NAS disk from Time Machine. Changed NAS from SMB 3 to SMB 2.1 max and turned off SMB signing on iMac. Restarted iMac. Connected to NAS in Finder fine. Select Disk in Time Machine does not show NAS however.

Disable multi-user backup toggle in time machine settings. Stop/start time machine.

Were you talking about doing so in QNAP? i didn't see these options. And My understanding of time machine in Mac OS is that there is not multiuser toggle. TM is only on or off and the user logged in at a particular point in time has no bearing on TM. TM backs up everything with permissions that upon restore the user only has acces to what they had before. There is no way to make a TM backup user specific when there are multiple users. What am I missing?
When I have SMB 2.1 on I cannot access the NAS volume through Time Machine preferences. When I switch back to 3.0 I can back up to NAS.
Write times have some spikes to 35MB/s but generally terrible at <5MB/s and a lot more of the time at KB / s

Are you running backup station or Hybrid Backup Sync? In Hybrid Backup Sync there is that option for multi-user backup under the TM settings. This setting is more around running multiple TM backups (potentially with different users) for TM.
As for speed - it will depend on your QNAP, your iMac, how you have it connected (wireless vs. wifi), etc.

This is what I followed to get TM running. I am not running either as I think the idea is to let TM do the work. It is the multiuser approach that I am taking because I have 4 macs to backup. Would I benefit from doing it different, like hybrid Backup Sync? Would that make it more fault tolerant of interruptions?

Speed seems bad. 460GB in 9.5 hours or about 14 MB per sec. But TM is notoriously slow on a first pass. Wired Ethernet connection through a Time Capsule used only as a switch.
TM ran most of the night and then crashed around 4:30 near as I can tell. It left my TM user connected to NAS so I had to restart NAS to clear it. Otherwise I have to rebuild the Mac SMB share completely I have learned. I assume it was due to Server Connection Interruption. I restarted it as you can tell from above and am just waiting for that failure. I have console running and will try to find the error when TM fails.

Hybrid Backup is an app I will use for sure. I'm not sure how it will handle multiple users. Does that mean multiple macs (user per mac)? It seems the macs would all use the same username (TimeMachine hardcoded) and overwrite each other's files. But I'm new to NAS so maybe QNAP has thought this through.
For those who come after me, my speed on TM is normal according to Pondini.org (http://pondini.org/TM/29.html). I am gettng ~48 GB per hour and I do have lots of small files.

Tried TM backup from different Mac running Mac OS 10.12.6. Same Server Connection Interruption error.

(QNAP) On SMB Advanced Config, I have async I/O UNCHECKED and SMB 2.1. I believe if you have multi-user backup checked then it sets it to SMB 3.0.
You might try disabling TM in HybridBackupSync (HBS). Then delete /share/CACHEDEV1_DATA/.timemachine directory. Then re-enable TM in HBS. Then try it again.
If this doesn't work, I would open a helpdesk ticket with QNAP on this.

I submited to helpdesk on the basis that these Server Connection Interruption errors are also occuring when Time Machine backups are disabled I am learning.
